GeneXP™ kits is Seegene’s original DSO™
-based multiplex
RT-PCR system.
- Only a single reaction in necessary to amplify multiple genes
of interest.
- One reaction is sufficient to amplify multiple genes simultaneously
in a single reaction.
GeneFishing™
DEG Dicovery kit uses randomly designed arbitrary primers to discover
DEGs (differentially expressed genes) in the samples, whereas the
GeneXp kit uses target specific primers for target gene profiling.
Multiplex PCRs capacity to simultaneously quantify miltiple mRNA
species from a single of RNA allows comparative analysis of different
mRNA species within samples. Variations in RNA isolation, initial
quantification errors or tube-to-tube variations in RT and PCR can
be compensated by including a known housekeeping gene. DSO™
primers can simultaneously detect multiple target genes without
creating any false products.

Comparison with other methods
To date, gene expression patterns are often analysed by microarray
methods that involve a DNA chip or by PCR-based methods.
Microarray
- Microarray analysis has the advantage over the other methods
in that it permits high throughput screening at one time. However,
it has many potential pitfalls, such as a high false-positive
rate and low sensitivity. Moreover, the number of samples that
can be compared is restricted, and it is difficult to detect subtle
differences between the samples.

- Experiments need to be repeated due to the low reproducibility.
- Hybridisation method is comparably lower in sensitivity than
PCR method.
- Additional experiments (Northern blot, Real-time PCR, RT-PCR,
etc.) are required for validation.
- All the clones with positive signal need to be checked, hence
requiring more cost and time for confirmation steps.
