Principle - Comparison of Primer Annealing
Three different primers are compared in the amplification of a
target sequence.
1) Comparison of primer
annealing |

Fig. 3. Comparison
of primer annealing 1. The flow chart of the general
PCR using three different primers: conventional short, conventional
longer, and ACP primers. Only ACP could generate a target
product. |

Fig. 4. Comparison of primer annealing 2. Amplification
of a target cDNA fragment using conventional primers and ACP.
The results shows that ACP generates a significant increase
in annealing specificity.

2) Sample result of
primer annealing |

Fig. 5. Result of primer
annealing. A pair of AC primers produced an expected
target product (lane 4). Conversely, a pair of conventional
longer primers generated many other non-specific products
including a target product (Lanes 2 and 3). |
